Requisition Number: ENSEM-2022-SB03
Scientist/Senior Scientist, Protein X-ray Crystallography
Shanghai Pudong, China
* ABOUT ENSEM THERAPEUTICS:
The name ENSEM connotes innovations in integrating groundbreaking technologies to decipher structural ENSEMbles and kinetics for drug design. A preclinical stage startup incubated and funded by the CBC group (http://www.cbridgecap.com), ENSEM focuses on high value and difficult-to-drug oncology targets with significant unmet medical needs. Its mission is to discover, develop, and commercialize transformative oncology therapies while leading the advancement of AI-driven structural ensemble and computational drug design.
* POSITION:
ENSEM Therapeutics is seeking an experienced and motivated PhD-level scientist specialized in protein X-ray crystallography to join our Shanghai-site Structure Biology team. In this role, you will manage as well as perform crystallographic experiments to help push forward our various structure-based drug design programs. Opportunities exist for additional contributions to other structural biology applications including NMR and CryoEM as well as various biophysical characterizations.
* POSITION DESCRIPTION:
§ Report to the Director of Structure Biology based in Boston and have the opportunities to manage research associates in Shanghai.
§ Manage our CRO collaborations on recombinant protein design, expression and purification, and protein X-ray crystallography. This includes reviewing the weekly data, troubleshooting, and planning for weekly and longer-term action items.
§ To participate in the downstream work of protein crystallography that includes refining small-molecule ligand-bound target complex structures and reporting/communicating the key structural findings to the team in writing and orally to aid in SBDD.
§ To engage assay development colleagues and medicinal and computational chemists to design assays, analyze compound SARs, and triage compounds for crystallographic studies.
§ To teamwork with internal colleagues to perform in-house crystallographic experiments ranging from protein production, crystallization, crystal manipulations, synchrotron data collection (at SSRF) and subsequent structure determination and interpretations.
§ To establish and manage crystal mail-in data collection services with foreign beamline scientists and/or CROs whenever additional/alternative beamtime is needed.
§ To provide project supports, serving as a structure biology champaign to efficiently accomplish project goals on time and budget; to conceive effective drugging strategies for established targets and provide critical assessments for new targets.
§ To contribute to platform development by monitoring and incorporating new technologies to enhance our protein sciences capabilities and SBDD supports; To help evaluate and elect platform tools to address specific program needs at a particular stage.
§ To contribute to the design and execution of relevant biophysical characterizations including but not limited to TSA, nanoDSF, ITC, SPR, and UVPD-MS.
* QUALIFICATIONS:
§ A PhD in Chemistry, Biochemistry, or Biophysics specialized in protein X-ray crystallography with 4+ years of relevant training and strong publications as well as solid understanding of protein chemistry and small-molecule drug discovery principles. Familiarity with NMR or CryoEM is a plus but not required.
§ Proficient in structural refinement and analysis to derive SBDD insights for med-chem designs as well as structure-mechanism hypotheses to help advance discovery projects; in protein design, expressions, purifications, and QCs; in protein/ligand co-crystallization, compound soaking, and cryoprotection. Experienced with in-person and mail-in synchrotron data collections.
§ Flexibility, adaptability, and multi-tasking in an interdisciplinary, hybrid (internal/outsourcing) and multi-site environment.
§ Exceptional interpersonal and communication skills to articulate deliverables, progresses, and issues to the drug discovery team and solicit feedbacks from team members and industrial and academic partners.
§ Personal attributes: attention to details while focused on the big picture; ability to scientifically influence and motivate others; a team player who listens effectively and values feedbacks; collaborative, open-minded, data-driven, and creative; persistent, optimistic with can-do spirit.
§ Exceptional Pharma/Biotech/CRO industrial veterans holding a MS degree and strong credentials are also encouraged to apply.
